Abstract
Design: Animal experiment study
Purpose: To evaluate the effect of MMC on conjunctival lymphatics and retinoic acid (EC 23) potential to induce lymphangiogenesis.
Method: 5 Sprague-Dawley rats were allocated in five groups (total 25) each were received either BSS, 0.1%MMC, 100nM EC 23, 100nM EC 23 with 0.1% MMC or 0.1% dimethyl sulfoxide in right eye. Left eyes were used as control. Eyes where enucleated at day 14 and each conjunctival specimen was stained with Masson trichome, CD 31 and D2-40. Fibrosis, lymphatic and vascular density was scored and compared within and between the groups.
Results: MMC has resulted significant reduction in lymphatic and conjunctival vascular density (p=0.009). 100nM EC 23 alone (p=.008) or combination with 0.1% MMC (p=.016) resulted in significant increase in lymphatic channel density score.
Conclusions: EC 23 has lymphangiogenetic property and shows biological action at 100 nM concentration.
